Blocking Agents Part 1 - 5/11/2009

Meridian Life Science, Inc. Blocking Agents Part 1

Do you have IgM detection interference? Meridian Life Science, Inc. has the solution:

Catalog #L15406G, Goat anti-Human IgG (Fc)

The validity of IgM detection can be compromised by the presence of pathogen-specific (immune) IgG.  There are two major mechanisms in which immune IgG can interfere with assays for IgM antibodies:

  • by competing with specific IgM for substrate binding sites
    • Immune IgG more effectively competes for available binding sites on an antigen substrate than does immune IgM.
    • In some instances, such as samples with low IgM titers, the competition by immune IgG can effectively block IgM binding and cause a false negative test result.
  • by forming immune complexes with Rheumatoid Factor (RF)

RF interference presents one of the most serious drawbacks to IgM testing.

Usually an IgM class, RF is an autoimmune antibody that can bind with immune IgG in the sample to form an immune complex which then can bind to the antigen.  This RF-IgG complex often cannot be distinguished from Immune IgM complexes and invariably yields an IgM false positive test result.
